This study aimed to explore the phytochemical profile and pharmacological effectiveness of ethanolic extract of Peperomia pellucida leaves on mice model of septic arthritis induced by Staphylococcus aureus. The study was conducted through two main stages: total flavonoid analysis and in vivo biological activity assay. Phytochemical analysis showed that the extract contains various bioactive compounds, including flavonoids (66.96 mg QE/g) that play a role in anti-inflammatory activity. The in vivo test involved five treatment groups of mice (n=5), including negative control, positive control (ciprofloxacin), and three graded doses of the extract (60, 80, 100 mg/kgBW). The parameters measured included distance traveled, movement speed, and gait pattern of mice. The results showed that a dose of 100 mg/kgBW was able to increase distance traveled and gait pattern scores significantly (p<0.05), while a dose of 60 mg/kgBW provided the highest speed increase. The Kruskal-Wallis and Mann-Whitney statistical tests confirmed the significance of differences between groups. These findings confirm that P. pellucida has potential as an effective phytotherapeutic agent in treating joint inflammation based on animal models
